Ivosidenib induces CYP3A4, and MMAE released from brentuximab vedotin is a CYP3A4 substrate. Lower MMAE exposure could theoretically reduce efficacy. Ivosidenib can also prolong the QT interval, but this is not directly relevant to this pairing.
What this interaction means in plain words
Brentuximab and Ivosidenib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Brentuximab belongs to Oncology; Ivosidenib belongs to Oncology.
Why it happens
CYP3A4 induction by ivosidenib may increase MMAE clearance.
What to do
Monitor for reduced response to brentuximab vedotin. Use an alternative if the loss of effect is a concern.
What to ask your pharmacist
- Should I take them at different times of day?
