Summary
Ceritinib prolongs the QT interval and is also a CYP3A4 inhibitor, which can raise pimozide concentrations. The combination substantially increases the risk of torsades de pointes.
What this interaction means in plain words
Ceritinib and Pimozide are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Ceritinib belongs to Oncology; Pimozide belongs to Central Nervous System.
Why it happens
Additive QT prolongation plus CYP3A4 inhibition reducing pimozide metabolism.
What to do
Avoid the combination and choose an alternative to pimozide during ceritinib therapy. If unavoidable, monitor ECG and electrolytes closely.
