Summary
Ceritinib can prolong the QT interval and cause bradycardia, and it inhibits CYP3A4, the main route of cisapride clearance. Cisapride exposure and QT risk can therefore both rise, risking torsades de pointes.
What this interaction means in plain words
Ceritinib and Cisapride are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Ceritinib belongs to Oncology; Cisapride belongs to Gastrointestinal Tract.
Why it happens
Additive QT prolongation plus CYP3A4 inhibition by ceritinib that raises cisapride concentrations.
What to do
Do not coadminister; stop cisapride and use an alternative prokinetic during ceritinib therapy.
What to ask your pharmacist
- Is there an alternative to one of these two medicines for me?
- If both are essential, who is monitoring me and how often?
- Which symptoms mean I should stop and seek help straight away?
