Summary

Ceritinib is a strong CYP3A4 inhibitor and may considerably raise eletriptan exposure. Elevated eletriptan levels increase the likelihood of vasospastic and hypertensive adverse effects.

What this interaction means in plain words

Ceritinib and Eletriptan are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Ceritinib belongs to Oncology; Eletriptan belongs to Central Nervous System.

Evidence level: Low

Why it happens

Ceritinib inhibits CYP3A4, reducing first-pass and systemic clearance of eletriptan.

What to do

Avoid eletriptan during ceritinib therapy and for 72 hours after a strong CYP3A4 inhibitor. Use a triptan not metabolized mainly by CYP3A4.
