Summary

Dabrafenib capsules dissolve less well at higher gastric pH, so acid suppression may lower its absorption and efficacy. Dabrafenib also induces CYP enzymes including CYP2C19 and CYP3A4, which can lower omeprazole exposure and reduce acid suppression.

What this interaction means in plain words

Dabrafenib and Omeprazole can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Dabrafenib belongs to Oncology; Omeprazole belongs to Gastrointestinal Tract.

Evidence level: Moderate

Why it happens

Omeprazole raises gastric pH, reducing dabrafenib solubility, while dabrafenib induces CYP2C19 and CYP3A4, increasing omeprazole clearance.

What to do

Monitor tumour response and reflux symptoms, and consider an alternative to the PPI such as a lower-impact acid suppressant if feasible. Increase omeprazole dose if acid control is inadequate.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
