Summary
Dacomitinib is a strong CYP2D6 inhibitor and venlafaxine is converted to its active metabolite O-desmethylvenlafaxine by CYP2D6. Inhibition raises venlafaxine and lowers the metabolite, while total active moiety changes little, but a rise in parent drug may increase adverse effects such as blood pressure elevation.
What this interaction means in plain words
Dacomitinib and Venlafaxine can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Dacomitinib belongs to Oncology; Venlafaxine belongs to Central Nervous System.
Why it happens
Inhibition of CYP2D6-mediated metabolism of venlafaxine.
What to do
Monitor blood pressure, heart rate and tolerability, and lower the venlafaxine dose if adverse effects emerge.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
