Summary

Imatinib inhibits CYP3A4, which forms the active metabolite of deflazacort. Exposure to the active metabolite rises, increasing corticosteroid adverse effects such as Cushingoid features, infection risk and adrenal suppression.

What this interaction means in plain words

Deflazacort and Imatinib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Deflazacort belongs to Miscellaneous; Imatinib belongs to Oncology.

Evidence level: Moderate

Why it happens

Imatinib inhibition of CYP3A4 reduces clearance of deflazacort's active metabolite.

What to do

Reduce deflazacort to about one-third of the usual dose when given with a moderate CYP3A4 inhibitor, as labeled. Monitor for corticosteroid toxicity.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
