Summary
Imatinib inhibits CYP3A4 and can substantially increase lomitapide exposure. This raises the risk of hepatotoxicity and gastrointestinal adverse effects. Lomitapide labeling contraindicates moderate and strong CYP3A4 inhibitors.
What this interaction means in plain words
Imatinib and Lomitapide are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Imatinib belongs to Oncology; Lomitapide belongs to Cardiovascular.
Why it happens
Imatinib inhibits CYP3A4, the main pathway of lomitapide metabolism, increasing its plasma concentrations.
What to do
Do not coadminister. Use an alternative lipid-lowering strategy while imatinib is required, or interrupt lomitapide if imatinib is unavoidable and follow labeling.
