Summary
Eliglustat can prolong cardiac conduction intervals at high concentrations, and donepezil can prolong QT and cause bradycardia. Combining them may increase arrhythmia risk, particularly when eliglustat exposure is increased by CYP2D6 or CYP3A inhibitors. Donepezil is also a CYP2D6 and CYP3A4 substrate.
What this interaction means in plain words
Donepezil and Eliglustat can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Donepezil belongs to Central Nervous System; Eliglustat belongs to Miscellaneous.
Why it happens
Additive QT and conduction effects.
What to do
Obtain a baseline ECG and correct electrolyte disturbances. Avoid additional QT-prolonging drugs or CYP inhibitors.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
