Summary
Itraconazole strongly increases eliglustat exposure, and both can prolong the QT interval. This raises the risk of cardiac conduction abnormalities and arrhythmia.
What this interaction means in plain words
Eliglustat and Itraconazole are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Eliglustat belongs to Miscellaneous; Itraconazole belongs to Anti Fungals.
Why it happens
Strong CYP3A4 inhibition raises eliglustat levels, with additive QT prolongation.
What to do
Avoid the combination, and especially so in CYP2D6 poor or intermediate metabolizers. Consider an alternative antifungal.
