Amiodarone and Eliglustat: contraindicated interaction.

For healthcare professionals — reference only. This page is a brief summary, not medical advice and not a substitute for official labeling or clinical judgement. Always verify before prescribing or dispensing.

Summary

Eliglustat is metabolized by CYP2D6 and CYP3A4, and amiodarone inhibits CYP2D6 and to some degree CYP3A4 and P-gp. Eliglustat exposure rises and may prolong PR, QRS and QT intervals. Both drugs also affect cardiac conduction.

What this interaction means in plain words

Amiodarone and Eliglustat are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Amiodarone belongs to Cardiovascular; Eliglustat belongs to Miscellaneous.

Evidence level: Moderate

Why it happens

Amiodarone inhibits CYP2D6 and CYP3A4, raising eliglustat levels, with additive conduction effects.

What to do

Avoid the combination; labeling advises against eliglustat with class Ia and III antiarrhythmics. Use an alternative antiarrhythmic if eliglustat is needed.
