Summary

Encorafenib affects CYP3A4 in both directions and inhibits OATP1B1, OATP1B3 and BCRP. Lovastatin exposure may therefore change unpredictably, with a risk of myopathy if levels rise or loss of efficacy if they fall.

What this interaction means in plain words

Encorafenib and Lovastatin can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Encorafenib belongs to Oncology; Lovastatin belongs to Cardiovascular.

Evidence level: Low

Why it happens

Encorafenib inhibits and induces CYP3A4 and inhibits hepatic uptake and efflux transporters involved in statin disposition.

What to do

Monitor for muscle symptoms and CK, and review lipid response; consider a different statin less dependent on CYP3A4 if problems arise.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
