Summary
Itraconazole is a strong CYP3A4 inhibitor and can increase estazolam concentrations. This may cause profound sedation, respiratory depression and prolonged impairment.
What this interaction means in plain words
Estazolam and Itraconazole are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Why it happens
Strong CYP3A4 inhibition by itraconazole reduces estazolam clearance.
What to do
Avoid the combination. Use a non-CYP3A4-dependent hypnotic such as temazepam or lorazepam if needed.
