Summary

Fosamprenavir is converted to amprenavir, a CYP3A4 inhibitor, and the effect is increased when boosted with ritonavir. Lovastatin exposure rises, increasing the risk of myopathy and rhabdomyolysis, and concurrent use is contraindicated.

What this interaction means in plain words

Fosamprenavir and Lovastatin are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Evidence level: High

Why it happens

Amprenavir, with or without ritonavir, inhibits CYP3A4, blocking lovastatin metabolism.

What to do

Do not combine. Use pravastatin or rosuvastatin, starting low, as an alternative.
