Summary
Fosamprenavir (as amprenavir, often boosted with ritonavir) inhibits CYP3A4 and can raise lurasidone concentrations substantially. The higher exposure increases the risk of dose-related adverse effects such as extrapyramidal symptoms and sedation. Labeling contraindicates strong CYP3A4 inhibitors with lurasidone.
What this interaction means in plain words
Fosamprenavir and Lurasidone are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Why it happens
Inhibition of CYP3A4-mediated metabolism of lurasidone.
What to do
Do not combine; select an alternative antipsychotic or antiretroviral regimen.
