Summary

Ribociclib prolongs the QT interval in a concentration-dependent manner, and gemtuzumab ozogamicin has also been associated with QT prolongation. Using both together may raise the risk of torsades de pointes, particularly if electrolyte disturbances are present.

What this interaction means in plain words

Gemtuzumab and Ribociclib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Evidence level: Low

Why it happens

Additive pharmacodynamic QT interval prolongation.

What to do

Obtain a baseline ECG and correct potassium and magnesium before starting, then repeat ECGs and electrolytes during co-treatment. Interrupt or reduce ribociclib if QTc becomes markedly prolonged.
