Summary

Ribociclib prolongs QT in a dose-dependent manner, and dronedarone adds to this effect. Ribociclib is also a CYP3A4 inhibitor and may raise dronedarone exposure, further increasing arrhythmia risk.

What this interaction means in plain words

Dronedarone and Ribociclib are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Dronedarone belongs to Cardiovascular; Ribociclib belongs to Oncology.

Evidence level: Moderate

Why it happens

Additive QT prolongation combined with CYP3A4 inhibition by ribociclib increasing dronedarone levels.

What to do

Do not combine; use an alternative antiarrhythmic or a different CDK4/6 inhibitor with lower QT risk.
