What this interaction means in plain words

Glecaprevir / pibrentasvir and Pravastatin can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Evidence level: High

Why it happens

Inhibition of hepatic uptake transporters (OATP1B1/1B3) and possibly BCRP by glecaprevir and pibrentasvir reduces pravastatin clearance.

What to do

Reduce the pravastatin dose by about half while the HCV regimen is given. Counsel the patient to report unexplained muscle pain or weakness, and check CK if symptoms occur.
