Summary
Lixisenatide improves glucose-dependent insulin secretion and, when added to a sulfonylurea such as glyburide, raises the risk of hypoglycemia. Glyburide's own insulin-releasing effect is independent of glucose, so the additive effect can be clinically important. Delayed gastric emptying from lixisenatide may also change the timing of glucose excursions.
What this interaction means in plain words
Glibenclamide and Lixisenatide can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Glibenclamide belongs to Endocrine; Lixisenatide belongs to Endocrine.
Why it happens
Additive pharmacodynamic effect on insulin secretion from a GLP-1 receptor agonist and a sulfonylurea.
What to do
Consider lowering the glyburide dose when lixisenatide is started. Monitor blood glucose and counsel the patient on recognising hypoglycemia.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
