Summary
Nefazodone is a strong CYP3A4 inhibitor and can considerably increase lurasidone exposure. This raises the risk of dose-related adverse effects including sedation and extrapyramidal symptoms. Strong CYP3A4 inhibitors are contraindicated with lurasidone.
What this interaction means in plain words
Lurasidone and Nefazodone are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Why it happens
Inhibition of CYP3A4-mediated metabolism of lurasidone.
What to do
Avoid the combination; use a different antidepressant that does not strongly inhibit CYP3A4.
