Summary
Saquinavir, usually boosted with ritonavir, strongly inhibits CYP3A4 and is expected to increase lurasidone concentrations markedly. This raises the risk of extrapyramidal symptoms, sedation and other dose-related effects. QT effects of saquinavir add further concern.
What this interaction means in plain words
Lurasidone and Saquinavir are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Why it happens
Inhibition of CYP3A4-mediated metabolism of lurasidone.
What to do
Do not coadminister; use an alternative antipsychotic or antiretroviral regimen.
