Summary

Opioids with serotonergic activity combined with MAO inhibitors have been associated with serotonin syndrome, and opioid labeling generally warns against MAOI use. Safinamide is a selective MAO-B inhibitor, which lowers the risk at recommended doses, but oliceridine may still add to sedation and respiratory depression.

What this interaction means in plain words

Oliceridine and Safinamide can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Oliceridine belongs to Musculoskeletal; Safinamide belongs to Central Nervous System.

Evidence level: Low

Why it happens

MAO inhibition with possible serotonergic opioid effects and additive CNS depression.

What to do

Monitor for agitation, tremor, hyperthermia, clonus and for sedation or respiratory depression, particularly after dose changes. Choose an alternative analgesic if signs of serotonin toxicity occur.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
