Both drugs can prolong the QT interval, so combined use increases the risk of serious ventricular arrhythmias including torsades de pointes. Osimertinib is also a weak CYP3A4 inducer that could slightly reduce pimozide exposure.
What this interaction means in plain words
Osimertinib and Pimozide can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Osimertinib belongs to Oncology; Pimozide belongs to Central Nervous System.
Why it happens
Additive QT interval prolongation.
What to do
Avoid the combination if possible, or monitor ECG and electrolytes periodically and correct hypokalemia and hypomagnesemia. Discontinue osimertinib if QTc becomes markedly prolonged.
