Summary
Ribociclib prolongs the QT interval and inhibits CYP3A4, which can increase pimozide exposure. Together they substantially increase the risk of torsades de pointes.
What this interaction means in plain words
Pimozide and Ribociclib are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Why it happens
Additive QT prolongation plus CYP3A4 inhibition by ribociclib reducing pimozide metabolism.
What to do
Do not co-administer; use an alternative to pimozide during ribociclib therapy.
