Summary

Glucocorticoids can blunt the growth-promoting effect of growth hormone. Growth hormone also inhibits 11-beta-HSD1, which reduces conversion of cortisone and prednisone to active cortisol and prednisolone, so glucocorticoid replacement may become inadequate.

What this interaction means in plain words

Prednisone and Somapacitan can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Prednisone belongs to Endocrine; Somapacitan belongs to Endocrine.

Evidence level: Moderate

Why it happens

Pharmacodynamic antagonism of growth hormone by glucocorticoids, and growth hormone inhibition of 11-beta-HSD1 that reduces activation of prednisone and cortisone.

What to do

Monitor growth and adrenal status, and adjust glucocorticoid doses as needed after starting somapacitan. Use the lowest effective glucocorticoid dose in growing children.
