Mécanisme d'action
Pentostatin is a potent transition state inhibitor of adenosine deaminase (ADA). ADA inhibition leads to cytotoxicity, believed to be due to elevated intracellular dATP, which can block DNA synthesis through inhibition of ribonucleotide reductase.

Mises en garde encadrées
- Give under supervision of a physician experienced in cancer chemotherapy. Higher doses than recommended are not advised; doses of 20-50 mg/m2 in divided doses over 5 days caused severe renal, liver, pulmonary and CNS toxicities. With fludarabine phosphate, 4 of 6 patients had severe or fatal pulmonary toxicity; the combination is not recommended.
Contre-indications
- Hypersensitivity to pentostatin (NIPENT).
Précautions particulières
