Summary

Inducers of CYP3A4 such as barbiturates lower isavuconazole concentrations considerably. Subtherapeutic exposure risks failure of treatment of serious invasive fungal infection. Strong inducers are contraindicated in the labeling.

What this interaction means in plain words

Amobarbital and Isavuconazonium are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Amobarbital belongs to Central Nervous System; Isavuconazonium belongs to Anti Fungals.

Evidence level: Moderate

Why it happens

Barbiturate-mediated CYP3A4 induction increases isavuconazole clearance.

What to do

Avoid the combination and choose a non-inducing sedative. If an inducer is essential, consider a different antifungal.
