Summary
Lonafarnib is a sensitive CYP3A4 substrate, so enzyme induction by amobarbital can greatly reduce its exposure. This could cause loss of efficacy. Labeling prohibits use with strong or moderate CYP3A inducers.
What this interaction means in plain words
Amobarbital and Lonafarnib are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Amobarbital belongs to Central Nervous System; Lonafarnib belongs to Miscellaneous.
Why it happens
Barbiturate-mediated CYP3A4 induction increases lonafarnib metabolism.
What to do
Avoid the combination. Select a non-inducing alternative to amobarbital.
