Summary

Barbiturates induce CYP3A4 and can substantially lower doravirine concentrations. This risks loss of virologic suppression and development of resistance. Doravirine labeling advises against strong CYP3A inducers.

What this interaction means in plain words

Amobarbital and Doravirine are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Amobarbital belongs to Central Nervous System; Doravirine belongs to Anti Virals.

Evidence level: Moderate

Why it happens

Barbiturate-mediated CYP3A4 induction increases doravirine metabolism.

What to do

Avoid the combination and use a non-inducing sedative. If an inducer is unavoidable, use an alternative antiretroviral regimen.

What to ask your pharmacist

  • Is there an alternative to one of these two medicines for me?
  • If both are essential, who is monitoring me and how often?
  • Which symptoms mean I should stop and seek help straight away?
