Summary
Ritonavir-boosted tipranavir inhibits CYP3A4 and markedly raises atorvastatin exposure. This increases the risk of myopathy and rhabdomyolysis, and labeling advises against the combination.
What this interaction means in plain words
Atorvastatin and Tipranavir are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Atorvastatin belongs to Cardiovascular; Tipranavir belongs to Anti Virals.
Why it happens
Inhibition of CYP3A4-mediated atorvastatin metabolism by ritonavir-boosted tipranavir.
What to do
Avoid atorvastatin with tipranavir/ritonavir; use an alternative statin such as pravastatin or rosuvastatin with dose limits. Monitor for muscle symptoms if any statin is used.
What to ask your pharmacist
- Is there an alternative to one of these two medicines for me?
- If both are essential, who is monitoring me and how often?
- Which symptoms mean I should stop and seek help straight away?
