Summary

Conivaptan is a strong CYP3A4 inhibitor and can markedly raise avanafil levels. Excess PDE5 inhibition may cause severe hypotension, syncope, visual changes and priapism.

What this interaction means in plain words

Avanafil and Conivaptan are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Evidence level: Moderate

Why it happens

CYP3A4 inhibition by conivaptan decreases avanafil clearance.

What to do

Avoid avanafil during conivaptan therapy. Use an alternative erectile dysfunction treatment if needed.
