Summary
Berotralstat inhibits CYP3A4 and may raise carbamazepine levels, risking dizziness, ataxia, diplopia and other toxicity. Carbamazepine, as a P-gp and CYP3A inducer, may also lower berotralstat exposure and reduce its prophylactic efficacy.
What this interaction means in plain words
Berotralstat and Carbamazepine can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Berotralstat belongs to Miscellaneous; Carbamazepine belongs to Central Nervous System.
Why it happens
Berotralstat inhibits CYP3A4 while carbamazepine induces CYP3A4 and P-glycoprotein, so each alters the other's exposure.
What to do
Monitor carbamazepine serum concentrations and signs of toxicity, and adjust the dose as needed. Monitor hereditary angioedema attack frequency for loss of berotralstat efficacy.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
