Summary
Bortezomib is metabolized partly by CYP3A4, and erythromycin may modestly increase its exposure. This could raise the risk of neuropathy, cytopenias and gastrointestinal toxicity.
What this interaction means in plain words
Bortezomib and Erythromycin can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Bortezomib belongs to Oncology; Erythromycin belongs to Anti Bacterial.
Why it happens
Inhibition of CYP3A4 by erythromycin reduces bortezomib clearance.
What to do
Monitor for neuropathy, thrombocytopenia and GI toxicity, and reduce the bortezomib dose if toxicity appears. Consider a non-inhibiting antibiotic.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
