Summary
Leniolisib inhibits OATP1B1 and OATP1B3, which mediate hepatic uptake of bosentan, so bosentan exposure may increase with greater hepatotoxicity and hypotension risk. Bosentan may also lower leniolisib exposure via CYP3A4 induction.
What this interaction means in plain words
Bosentan and Leniolisib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Bosentan belongs to Miscellaneous; Leniolisib belongs to Miscellaneous.
Why it happens
Leniolisib inhibits OATP1B1/1B3 uptake of bosentan, and bosentan induces CYP3A4 metabolism of leniolisib.
What to do
Monitor liver enzymes and blood pressure, and assess clinical response to leniolisib. Consider alternatives if either effect becomes significant.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
