Summary
Crizotinib is a moderate CYP3A4 inhibitor and lomitapide is a sensitive CYP3A4 substrate, so exposure to lomitapide can rise markedly. This increases the risk of hepatotoxicity and gastrointestinal adverse effects.
What this interaction means in plain words
Crizotinib and Lomitapide are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Crizotinib belongs to Oncology; Lomitapide belongs to Cardiovascular.
Why it happens
Crizotinib inhibits CYP3A4, reducing first-pass and systemic metabolism of lomitapide.
What to do
Avoid the combination; labeling for lomitapide advises against moderate and strong CYP3A4 inhibitors. Use an alternative lipid-lowering strategy during crizotinib treatment.
