Summary

Tafenoquine inhibits renal cation transporters in vitro, which can reduce metformin clearance and raise its plasma exposure. Higher metformin levels increase the risk of gastrointestinal intolerance and, in susceptible patients, lactic acidosis.

What this interaction means in plain words

Dapagliflozin-metformin and Tafenoquine can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Dapagliflozin-metformin belongs to Endocrine; Tafenoquine belongs to Anti Parasitic.

Evidence level: Moderate

Why it happens

Tafenoquine inhibits OCT2 and MATE transporters, reducing renal tubular secretion of metformin.

What to do

Avoid coadministration where possible; if tafenoquine is required, monitor glucose, renal function and signs of metformin toxicity. Consider reducing the metformin dose if adverse effects appear.
