Summary
Dronedarone inhibits P-glycoprotein and is a moderate CYP3A4 inhibitor, and docetaxel is cleared mainly by CYP3A4. Exposure to docetaxel may rise, increasing myelosuppression, neutropenia, fluid retention and neurotoxicity.
What this interaction means in plain words
Docetaxel and Dronedarone can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Docetaxel belongs to Oncology; Dronedarone belongs to Cardiovascular.
Why it happens
Inhibition of CYP3A4-mediated metabolism and P-glycoprotein efflux by dronedarone reduces docetaxel clearance.
What to do
Monitor blood counts and signs of docetaxel toxicity closely, and consider dose reduction if adverse effects emerge. Consider an alternative antiarrhythmic if feasible.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
