Summary

Ritonavir is a potent CYP3A4 inhibitor and markedly raises dronedarone levels, increasing the risk of QT prolongation and torsades de pointes. Dronedarone may also modestly raise ritonavir exposure through P-glycoprotein inhibition.

What this interaction means in plain words

Dronedarone and Ritonavir are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Dronedarone belongs to Cardiovascular; Ritonavir belongs to Anti Virals.

Evidence level: High

Why it happens

Potent CYP3A4 inhibition by ritonavir reduces dronedarone clearance, with additional P-glycoprotein inhibition by dronedarone.

What to do

Do not coadminister; select an alternative antiarrhythmic or antiretroviral regimen.

What to ask your pharmacist
