Summary

Nefazodone is a potent CYP3A4 inhibitor and can markedly increase eletriptan exposure. This raises the risk of vasospasm, hypertension and other triptan toxicity.

What this interaction means in plain words

Eletriptan and Nefazodone are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Evidence level: Moderate

Why it happens

Inhibition of CYP3A4 metabolism of eletriptan.

What to do

Avoid eletriptan within 72 hours of nefazodone. Use a triptan not dependent on CYP3A4 if needed.
