Summary
Cobicistat is a strong CYP3A4 inhibitor and markedly raises eletriptan exposure, which increases the risk of triptan toxicity including vasoconstriction and hypertension. Eletriptan labeling contraindicates use with strong CYP3A4 inhibitors.
What this interaction means in plain words
Cobicistat and Eletriptan are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Cobicistat belongs to Miscellaneous; Eletriptan belongs to Central Nervous System.
Why it happens
Inhibition of CYP3A4-mediated metabolism of eletriptan, increasing its plasma concentrations.
What to do
Do not use eletriptan during or within about 72 hours of cobicistat. Use a triptan not dependent on CYP3A4, such as rizatriptan or sumatriptan, if one is needed.
