Summary
Eslicarbazepine may reduce lorlatinib exposure through CYP3A induction. Strong inducers raise the risk of hepatotoxicity as well as reduced efficacy, so the combination is best avoided.
What this interaction means in plain words
Eslicarbazepine and Lorlatinib can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Eslicarbazepine belongs to Central Nervous System; Lorlatinib belongs to Oncology.
Why it happens
CYP3A4 induction by eslicarbazepine increases lorlatinib metabolism.
What to do
Avoid combined use and choose a non-inducing antiseizure drug. If unavoidable, monitor liver enzymes, bilirubin and tumor response.
What to ask your pharmacist
- Does the benefit of taking both outweigh the risk in my case?
- Do I need any blood test, ECG or other check while on both?
