What this interaction means in plain words

Glasdegib and Itraconazole can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.

Glasdegib belongs to Oncology; Itraconazole belongs to Anti Fungals.

Evidence level: Moderate

Why it happens

Additive QT prolongation, plus itraconazole inhibition of CYP3A4-mediated glasdegib metabolism.

What to do

Use an alternative antifungal where possible. If the combination is unavoidable, obtain a baseline ECG, monitor ECG and electrolytes, and consider reducing the glasdegib dose.
