Itraconazole can increase palovarotene exposure by inhibiting its metabolism. This may amplify retinoid toxicity such as mucocutaneous effects, premature growth plate closure in children and hypervitaminosis A-like effects.
What this interaction means in plain words
Itraconazole and Palovarotene can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Itraconazole belongs to Anti Fungals; Palovarotene belongs to Miscellaneous.
Why it happens
Strong CYP3A4 inhibition reduces palovarotene clearance.
What to do
Avoid concurrent use of strong CYP3A4 inhibitors with palovarotene; select a non-interacting antifungal. If unavoidable, monitor closely for retinoid toxicity.
