Summary

Both drugs prolong the QT interval, and ribociclib may also raise vandetanib levels through CYP3A4 inhibition. Together this raises the risk of torsades de pointes and sudden death.

What this interaction means in plain words

Ribociclib and Vandetanib are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Ribociclib belongs to Oncology; Vandetanib belongs to Oncology.

Evidence level: Moderate

Why it happens

Additive QT prolongation, with possible increased vandetanib exposure from CYP3A4 inhibition by ribociclib.

What to do

Avoid the combination; use an alternative that does not prolong QT. Vandetanib labeling warns against combining it with other QT-prolonging drugs.
