Mécanisme d'action
Temporarily replaces the missing Factor X needed for effective hemostasis. Factor X is activated to Factor Xa, which with Factor Va forms the prothrombinase complex that converts prothrombin to thrombin, leading to a cross-linked fibrin clot.
Posologie adulte habituelle (résumé de la notice)
Routine prophylaxis of bleeding episodes (adults and adolescents 12 years or older): 25 IU/kg IV twice weekly; adjust to clinical response; do not administer more than 60 IU/kg daily
Grossesse et allaitement
- Grossesse
- Unknown Limited human data and no animal studies. Factor X deficiency itself raises the risk of bleeding and miscarriage, so use when clearly needed under a specialist.
- Allaitement
- Unknown No data on milk levels or effects on the infant. A large plasma protein, unlikely to be absorbed by the infant. Weigh the benefits of breastfeeding against the mother's need for treatment.
Effets indésirables signalés
- Infusion site erythema
- Infusion site pain
- Fatigue
- Back pain
Contre-indications
- Life-threatening hypersensitivity reactions to COAGADEX
Précautions particulières
- Hypersensitivity reactions including anaphylaxis are possible; discontinue and treat if symptoms occur
- Neutralizing antibodies (inhibitors) may develop; if expected Factor X levels are not attained or bleeding is uncontrolled, assay for inhibitor
- Made from human blood; may transmit infectious agents including viruses and vCJD/CJD agents
