Indications
Mécanisme d'action
Orally bioavailable small molecule TPO receptor agonist that stimulates proliferation and differentiation of megakaryocytes from bone marrow progenitor cells, increasing platelet production. It does not compete with TPO for receptor binding and has an additive effect with TPO.
Posologie adulte habituelle (résumé de la notice)
Thrombocytopenia in chronic liver disease before a procedure (tablets): platelet count less than 40×10 9 /L: 60 mg (3 tablets) orally once daily for 5 days, starting 10 to 13 days before the procedure
Grossesse et allaitement

- Unknown Animal studies showed miscarriage in rabbits and pup deaths in rats at high exposures; human data are insufficient. May cause fetal harm. Use only if benefit outweighs risk and inform the patient of the potential risk.
- Allaitement
- Unsafe No human milk data; present in rat milk. US label: do not breastfeed during treatment and for at least 2 weeks after the last dose; after a short pre-procedure course, pump and discard milk for that period.

Contre-indications
- None.
Précautions particulières
- Thrombotic/thromboembolic complications: TPO receptor agonists are associated with thrombotic and thromboembolic events; monitor platelet counts and for signs of thromboembolism, and treat promptly.
- Consider increased thrombotic risk in patients with known risk factors, including genetic prothrombotic conditions and acquired factors such as antiphospholipid syndrome.
- Do not use to normalize platelet counts.
