Mécanisme d'action
Vascular endothelial growth factor receptor 2 (VEGFR2) antagonist that specifically binds VEGF receptor 2 and blocks binding of VEGFR ligands, VEGF-A, VEGF-C, and VEGF-D. As a result, ramucirumab inhibits ligand-stimulated activation of VEGF2, thereby inhibiting ligand-induced proliferation, and migration of human endothelial cells.
Posologie adulte habituelle (résumé de la notice)
Non-Small Cell Lung Cancer: Ramucirumab 10 mg/kg IV on Day 1, PLUS Docetaxel 75 mg/m2 IV on Day 1, Repeat cycle every 21 days, Continue until disease progression or unacceptable toxicity.
Grossesse et allaitement
- Grossesse
- Unsafe Can cause fetal harm: it blocks VEGFR2, which the placenta and fetal blood vessels need (mice lacking VEGFR2 die as embryos); no human or animal reproduction studies. Check pregnancy status before starting; use effective contraception during treatment and for 3 months after the last dose.
- Allaitement
- Unsafe No human milk data. As a large antibody, milk levels are likely very low and poorly absorbed, but serious infant reactions are possible. Do not breastfeed during treatment and for 2 months after the last dose (US label; LactMed concurs with caution).

Précautions particulières
- Impaired wound healing can occur with antibodies inhibiting the VEGF or VEGFR pathway; VEGFR2 antagonist has the potential to adversely affect wound healing
- Withhold for 28 days prior to elective surgery; do not administer for at least 2 weeks following a major surgical procedure and until adequate wound healing
- May cause hypothyroidism; monitor thyroid function during treatment
