Mécanisme d'action
CD30-directed antibody-drug conjugate: a chimeric IgG1 linked to the microtubule-disrupting agent MMAE. After binding CD30-expressing cells and internalization, MMAE is released and binds tubulin, causing cell cycle arrest and apoptosis.
Posologie adulte habituelle (résumé de la notice)
Monotherapy for adults: 1.8 mg/kg up to a maximum of 180 mg, intravenous infusion over 30 minutes, every 3 weeks
Grossesse et allaitement
- Grossesse
- Unsafe Antibody-drug conjugate (cytotoxic MMAE): can cause fetal harm. Avoid in pregnancy. Women: effective contraception during treatment and for 2 months after; men with female partners: 4 months.
- Allaitement
- Unsafe No human milk data. Avoid breastfeeding during treatment because of possible serious infant harm (low blood counts, nerve or gut toxicity).

- JC virus infection resulting in progressive multifocal leukoencephalopathy (PML) and death can occur in patients receiving ADCETRIS.
Contre-indications
- Concomitant use with bleomycin due to pulmonary toxicity
Précautions particulières
- Peripheral neuropathy is cumulative, predominantly sensory; monitor and modify dose, delay or discontinue.
- Anaphylaxis and infusion reactions: monitor during infusion; permanently discontinue for anaphylaxis.
- Hematologic toxicities including fatal febrile neutropenia; monitor CBC before each dose; G-CSF primary prophylaxis in combination regimens.
