Summary

Phenobarbital is a long-acting barbiturate that depresses the CNS, so combining it with amisulpride may increase drowsiness, impair coordination and reduce alertness. Antipsychotics may also lower seizure threshold, which can counteract the anticonvulsant purpose of phenobarbital.

What this interaction means in plain words

Amisulpride and Phenobarbital can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Evidence level: Moderate

Why it happens

Additive CNS depression through GABA-A potentiation and the sedative properties of amisulpride.

What to do

Monitor sedation and seizure control, and start amisulpride at a low dose with gradual titration. Counsel patients to avoid alcohol and driving until the response is known.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
