Summary
Barbiturates induce CYP3A4 and can lower amlodipine concentrations, possibly weakening blood pressure control. Barbiturates may also add to orthostatic hypotension caused by the antihypertensive combination and by the diuretic.
What this interaction means in plain words
Amlodipine-hydrochlorothiazide-valsartan and Amobarbital can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Amlodipine-hydrochlorothiazide-valsartan belongs to Cardiovascular; Amobarbital belongs to Central Nervous System.
Why it happens
Induction of CYP3A4 by amobarbital increases amlodipine metabolism, with additive hypotensive and sedative effects.
What to do
Monitor blood pressure after starting or stopping amobarbital and increase the amlodipine dose if needed. Counsel patients about orthostatic dizziness.
